ThermoFisher Attune NxT Maintenance Guidelines

1. Log into windows with your RUNet login name and password
2. Double click “Attune NxT Software” on the desktop and login with your user name and password
3. Check the “Instrument” menu on the top. If the instrument is OFF, only “Startup” icon is active and all others are inactive. If the instrument is ON, please go to step 5
4. Select “Startup”. Follow the wizard’s guidance: unload the tube from the tube lifter and discard the tube; take out the 96-well cleaning plate from the auto-sampler and put on the top of the instrument. Select “next”. The whole procedure will take approximately 3 min
5. From the “Instrument” menu, select “Sanitize Attune SIP”. Follow the wizard’s guidance: load a tube of 3 ml of 10% bleach on the holder and raise the tube lifter. When it is done unload the tube and discard. Run “Sanitize Attune SIP” again with DI water. Discard the tube after it is done
6. From the main page, select “New Experiment” to start a new experiment; or “Import Experiment” to open an old experiment; or “New Experiment from a Template”
7. Run your experiment
8. When you are done with the experiment, export the experiment to “T:\your folder” and FCS files to “T:\FCS” folder
9. Copy your experiment and FCS files to your CFS for storage
10. Before you leave FCRC please do quick “Deep Clean” or switch instrument OFF depending on the time of the day:
    • During the daytime (business hours), please do quick “Deep Clean”
      • From the “Instrument” menu select “Deep Clean”.  Press the arrow to get to the submenu and select “Quick”
      • Follow the wizard’s guidance: load a tube of 3 ml of 10% bleach on the holder and raise the tube lifter. Rinse the first 3 wells of the 96-well cleaning plate and put it into the auto sampler. Select “next” from the “Deep Clean” wizard. The whole procedure will take approximately 30 min
    • If you finish experiment after 6 pm on weekday or any time in the weekend (after hours), please switch instrument OFF
      • From the “Instrument” menu select “Shutdown”. Press the arrow to get to the submenu and select “Thorough”
      • Follow the wizard’s guidance: load a tube of 3 ml of 10% bleach on the holder and raise the tube lifter. Rinse the first 3 wells of the 96-well cleaning plate and put it into the auto sampler. Select “next” from the “Shutdown” wizard. The whole procedure will take approximately 75 min
11. Log out your account and close the Attune software. You don’t have to wait until the “Quick Deep Clean” or “Shutdown” procedure finish
12. Please be sure to logoff Windows before you leave FCRC. Otherwise in 10 minutes Windows will automatically lock the session under your name and will prevent other researchers from usage of the instrument

After hours — e-mail to FCRC_Staff@rockefeller.edu; leave the note about the problem on the instrument’s keyboard; proceed on step 10-12 from the above list
