ATP-binding cassette (ABC) transporters, with more than 2000 members identified, constitute one of the largest protein superfamilies. They are integral to numerous biological processes and many are medically important. In this talk, Dr. Chen will first present crystal structures of the E. coli maltose importer captured in multiple conformations and discuss how the free energy of ATP hydrolysis is harnessed to transport maltose against its concentration gradient. She will also present structural studies of two ABC exporters, a eukaryotic multidrug transporter called P-glycoprotein (P-gp) and a bacterial polypeptide transporter of the type I secretion system (T1SS). Maltose transporter, P-gp and T1SS contain similar ATP-binding domains, which enable them to hydrolyze ATP and achieve active transport. They also exhibit unique transmembrane pathways that correlate beautifully with the very different physical and chemical properties of their respective substrates.
Dr. Chen received her bachelor’s of science in chemistry from Ohio University and her Ph.D. in biochemistry from Harvard University, where she was advised by Don C. Wiley. She did postdoctoral work at Baylor College of Medicine with Florante A. Quiocho. She joined Purdue University in 2002 as assistant professor of biology and was promoted to associate professor in 2007 and professor in 2011. She was named a Pew Scholar in 2003 and was named an investigator of the Howard Hughes Medical Institute in 2008. She joined Rockefeller’s faculty in 2014.
