Granulocyte Colony-Stimulating Factor Protects Mice during Respiratory Virus Infections

A burst in the production of pro-inflammatory molecules characterizes the beginning of the host response to infection. Cytokines, chemokines, and growth factors work in concert to control pathogen replication and activate innate and adaptive immune responses. Granulocyte colony-stimulating factor (G-CSF) mobilizes and activates hematopoietic cells from the bone marrow, and it has been shown to mediate the generation of effective immunity against bacterial and fungal infections. G-CSF is produced at high levels in the lungs during infection with influenza and parainfluenza viruses, but its role during these infections is unknown. Here we show that during infection of mice with a non-lethal dose of influenza or Sendai virus, G-CSF promotes the accumulation of activated Ly6G(+) granulocytes that control the extent of the lung pro-inflammatory response. Remarkably, these G-CSF-mediated effects facilitate viral clearance and sustain mouse survival.

Suppression of non-prompt J/psi, prompt J/psi, and Upsilon(1S) in PbPb collisions at root s(NN)=2.76 TeV

Yields of prompt and non-prompt J/psi ,as well as Upsilon(1S) mesons, are measured by the CMS experiment via their mu(+)mu(-) decays in PbPb and pp collisions at root s(NN) - 2.76TeV for quarkonium rapidity |y| < 2.4. Differential cross sections and nuclear modification factors are reported as functions of y and transverse momentum p(T), as well as collision centrality. For prompt J/psi with relatively high p(T) (6.5 < p(T) < 30 GeV/c), a strong, centrality-dependent suppression is observed in PbPb collisions, compared to the yield in pp collisions scaled by the number of inelastic nucleon-nucleon collisions. In the same kinematic range, a suppression of non-prompt J/psi, which is sensitive to the in-medium b-quark energy loss, is measured for the first time. Also the low-p(T) Upsilon(1S) mesons are suppressed in PbPb collisions.

Measurement of the masses and widths of the bottom baryons Sigma(+/-)(b) and Sigma*(+/-therefore)(b)

Using data from p (p) over bar collisions at root s = 1.96 TeV recorded by the CDF II detector at the Fermilab Tevatron, we present improved measurements of the masses and first measurements of natural widths of the four bottom baryon resonance states Sigma(+)(b), Sigma*(+)(b) and Sigma(-)(b), Sigma*(-)(b). These states are fully reconstructed in their decay modes to Lambda(0)(b)pi(+/-) where Lambda(0)(b) -> Lambda(+)(c)pi(-) with Lambda(+)(c) -> pK(-) pi(+). The analysis is based on a data sample corresponding to an integrated luminosity of 6.0 fb(-1) collected by an online event selection based on tracks displaced from the p (p) over bar interaction point.

An RNA interference screen uncovers a new molecule in stem cell self-renewal and long-term regeneration

Adult stem cells sustain tissue maintenance and regeneration throughout the lifetime of an animal(1,2). These cells often reside in specific signalling niches that orchestrate the stem cell's balancing act between quiescence and cell-cycle re-entry based on the demand for tissue regeneration(2-4). How stem cells maintain their capacity to replenish themselves after tissue regeneration is poorly understood. Here we use RNA-interference-based loss-of-function screening as a powerful approach to uncover transcriptional regulators that govern the self-renewal capacity and regenerative potential of stem cells. Hair follicle stem cells provide an ideal model. These cells have been purified and characterized from their native niche in vivo and, in contrast to their rapidly dividing progeny, they can be maintained and passaged long-term in vitro(5-7). Focusing on the nuclear proteins and/or transcription factors that are enriched in stem cells compared with their progeny(5,6), we screened similar to 2,000 short hairpin RNAs for their effect on long-term, but not short-term, stem cell self-renewal in vitro. To address the physiological relevance of our findings, we selected one candidate that was uncovered in the screen: TBX1. This transcription factor is expressed in many tissues but has not been studied in the context of stem cell biology. By conditionally ablating Tbx1 in vivo, we showed that during homeostasis, tissue regeneration occurs normally but is markedly delayed. We then devised an in vivo assay for stem cell replenishment and found that when challenged with repetitive rounds of regeneration, the Tbx1-deficient stem cell niche becomes progressively depleted. Addressing the mechanism of TBX1 action, we discovered that TBX1 acts as an intrinsic rheostat of BMP signalling: it is a gatekeeper that governs the transition between stem cell quiescence and proliferation in hair follicles. Our results validate the RNA interference screen and underscore its power in unearthing new molecules that govern stem cell self-renewal and tissue-regenerative potential.

Search for quark compositeness in dijet angular distributions from pp collisions at root s=7 TeV

A search for quark compositeness using dijet angular distributions from pp collisions at root s = 7TeV is presented. The search has been carried out using a data sample corresponding to an integrated luminosity of 2 : 2 fb(-1), recorded by the CMS experiment at the LHC. Normalized dijet angular distributions have been measured for dijet invariant masses from 0.4TeV to above 3TeV and compared with a variety of contact interaction models, including those which take into account the effects of next-to-leading-order QCD corrections. The data are found to be in agreement with the predictions of perturbative QCD, and lower limits are obtained on the contact interaction scale, ranging from 7.5 up to 14.5TeV at 95% confidence level.

Measurement of B-s(0) -> D-s(()*D-)+(s)(*()-) Branching Ratios

The decays B-s(0) -> D-s(()*D-)+(s)(*()-) are reconstructed in a data sample corresponding to an integrated luminosity of 6: 8 fb(-1) collected by the CDF II detector at the Tevatron p (p) over bar collider. All decay modes are observed with a significance of more than 10 sigma, and we measure the B-s(0) production rate times B-s(0) -> D-s(()*D-)+(s)(*()-) branching ratios relative to the normalization mode B-0 -> Ds+D- to be 0.183 +/- 0.021 +/- 0.017 for B-s(0) -> Ds+Ds-, 0.424 +/- 0.046 +/- 0.035 for B-s(0) -> D-s*D-+(s)-/+, 0.654 +/- 0.072 +/- 0.065 for B-s(0) -> D-s*D-+(s)*(-), and 1.261 +/- 0.095 +/- 0.112 for the inclusive decay B-s(0) -> D-s(()*D-)+(s)(*()-), where the uncertainties are statistical and systematic. These results are the most precise single measurements to date and provide important constraints for indirect searches for nonstandard model physics in B-s(0) mixing.

CEF1/CDC5 alleles modulate transitions between catalytic conformations of the spliceosome

Conformational change within the spliceosome is required between the first and second catalytic steps of pre-mRNA splicing. A prior genetic screen for suppressors of an intron mutant that stalls between the two steps yielded both prp8 and non-prp8 alleles that suppressed second-step splicing defects. We have now identified the strongest non-prp8 suppressors as alleles of the NTC (Prp19 complex) component, CEF1. These cef1 alleles generally suppress second-step defects caused by a variety of intron mutations, mutations in U6 snRNA, or deletion of the second-step protein factor Prp17, and they can activate alternative 39 splice sites. Genetic and functional interactions between cef1 and prp8 alleles suggest that they modulate the same event(s) in the first-to-second-step transition, most likely by stabilization of the second-step spliceosome; in contrast, alleles of U6 snRNA that also alter this transition modulate a distinct event, most likely by stabilization of the first-step spliceosome. These results implicate a myb-like domain of Cef1/CDC5 in interactions that modulate conformational states of the spliceosome and suggest that alteration of these events affects splice site use, resulting in alternative splicing-like patterns in yeast.

Staphylococcus epidermidis Colonization Is Highly Clonal Across US Cardiac Centers

Background. Little is known about the clonality of Staphylococcus epidermidis in the United States, although it is the predominant pathogen in infections involving prosthetic materials, including ventricular assist devices (VADs). Methods. Seventy-five VAD recipients at 4 geographically diverse US cardiac centers were prospectively followed up to 1 year of VAD support. The anterior nares, sternum, and (future) driveline exit site were cultured for S. epidermidis before VAD insertion and at 7 times after surgery. Infection isolates were also collected. Isolates were typed by pulsed-field gel electrophoresis. A subset underwent susceptibility testing and staphylococcal chromosomal cassette mec and multilocus sequence typing. Results. A total of 1559 cultures yielded 565 S. epidermidis isolates; 254 of 548 typed isolates (46%) belonged to 1 of 7 clonal types as defined by pulsed-field gel electrophoresis. These clones were identified in up to 27 people distributed across all 4 cardiac centers. They caused 3 of 6 VAD-related infections. Disseminated clones were more antibiotic resistant than were less prevalent isolates (eg, 79% vs 54% methicillin resistant; P = .0021). Conclusions. This study revealed that healthcare-associated S. epidermidis infection is remarkably clonal. We describe S. epidermidis clones that are highly resistant to antibiotics distributed across US cardiac centers. A High-Fat Diet Is Associated With Endotoxemia That Originates From the Gut

Endotoxemia, characterized by an excess of circulating bacterial wall lipopolysaccharide, is associated with systemic inflammation and the metabolic syndrome. Placing 8 healthy subjects on a Western-style diet for 1 month induced a 71% increase in plasma levels of endotoxin activity (endotoxemia), whereas a prudent-style diet reduced levels by 31%. The Western-style diet might, therefore, contribute to endotoxemia by causing changes in gastrointestinal barrier function or the composition of the microbiota. Endotoxemia might also develop in individuals with gastrointestinal barrier impairment. Therapeutic reagents that reduce endotoxemia might reduce systemic inflammation in patients with gastrointestinal diseases or metabolic syndrome.

Genome-Wide Analysis of GLD-1-Mediated mRNA Regulation Suggests a Role in mRNA Storage

Translational repression is often accompanied by mRNA degradation. In contrast, many mRNAs in germ cells and neurons are "stored" in the cytoplasm in a repressed but stable form. Unlike repression, the stabilization of these mRNAs is surprisingly little understood. A key player in Caenorhabditis elegans germ cell development is the STAR domain protein GLD-1. By genome-wide analysis of mRNA regulation in the germline, we observed that GLD-1 has a widespread role in repressing translation but, importantly, also in stabilizing a sub-population of its mRNA targets. Additionally, these mRNAs appear to be stabilized by the DDX6-like RNA helicase CGH-1, which is a conserved component of germ granules and processing bodies. Because many GLD-1 and CGH-1 stabilized mRNAs encode factors important for the oocyte-to-embryo transition (OET), our findings suggest that the regulation by GLD-1 and CGH-1 serves two purposes. Firstly, GLD-1-dependent repression prevents precocious translation of OET-promoting mRNAs. Secondly, GLD-1- and CGH-1-dependent stabilization ensures that these mRNAs are sufficiently abundant for robust translation when activated during OET. In the absence of this protective mechanism, the accumulation of OET-promoting mRNAs, and consequently the oocyte-to-embryo transition, might be compromised.
