The molecular circuitry underlying pluripotency in embryonic stem cells

Cells in the pluripotent state have the ability to self-renew indefinitely and to differentiate to all the cells of the embryo. These cells provide an in vitro window into development, including human development, as well as holding extraordinary promise for cell-based therapies in regenerative medicine. The recent demonstration that somatic cells can be reprogrammed to the pluripotent state has raised the possibility of patient and disease-specific induced pluripotent cells. In this article, we review the molecular underpinning of pluripotency. We focus on the transcriptional and signaling networks that underlie the state of pluripotency and control differentiation. In general, the action of each of the molecular components and pathways is dose and context dependent highlighting the need for a systems approach to understanding pluripotency. Immunity to infection in IL-17-deficient mice and humans

Mice with defective IL-17 immunity display a broad vulnerability to various infectious agents at diverse mucocutaneous surfaces. In humans, the study of patients with various primary immunodeficiencies, including autosomal dominant hyper-IgE syndrome caused by dominant-negative STAT3 mutations and autosomal recessive autoimmune polyendocrinopathy syndrome type 1 caused by null mutations in AIRE, has suggested that IL-17A, IL-17F and/or IL-22 are essential for mucocutaneous immunity to Candida albicans. This hypothesis was confirmed by the identification of rare patients with chronic mucocutaneous candidiasis (CMC) due to autosomal recessive IL-17RA deficiency and autosomal dominant IL-17F deficiency. Heterozygosity for gain-of-function mutations in STAT1 in additional patients with CMC was recently shown to inhibit the development of IL-17 T cells. Although the infectious phenotype of patients with CMC and inborn errors of IL-17 immunity remains to be finely delineated, it appears that human IL-17A and IL-17F display redundancy for protective immunity in natural conditions that is not seen in their mouse orthologs in experimental conditions.

Allometric scaling of population variance with mean body size is predicted from Taylor's law and density-mass allometry

Two widely tested empirical patterns in ecology are combined here to predict how the variation of population density relates to the average body size of organisms. Taylor's law (TL) asserts that the variance of the population density of a set of populations is a power-law function of the mean population density. Density-mass allometry (DMA) asserts that the mean population density of a set of populations is a power-law function of the mean individual body mass. Combined, DMA and TL predict that the variance of the population density is a power-law function of mean individual body mass. We call this relationship "variance-mass allometry" (VMA). We confirmed the theoretically predicted power-law form and the theoretically predicted parameters of VMA, using detailed data on individual oak trees (Quercus spp.) of Black Rock Forest, Cornwall, New York. These results connect the variability of population density to the mean body mass of individuals.

Measurement of the Difference in CP-Violating Asymmetries in D-0 -> K+K- and D-0 -> pi(+)pi(-) Decays at CDF

We report a measurement of the difference (Delta A(CP)) between time-integrated CP-violating asymmetries in D-0 -> K+K- and D-0 -> pi(+)pi(-) decays reconstructed in the full data set of proton-antiproton collisions collected by the Collider Detector at Fermilab, corresponding to 9.7 fb(-1) of integrated luminosity. The strong decay D*(+) -> D-0 pi(+) is used to identify the charm meson at production as D-0 or (D) over bar (0). We measure Delta A(CP) = [-0.62 +/- 0.21(stat) +/- 0.10(syst)]%, which differs from zero by 2.7 Gaussian standard deviations. This result supports similar evidence for CP violation in charm-quark decays obtained in proton-proton collisions.

Search for high-mass resonances decaying into tau-lepton pairs in pp collisions at root s=7 TeV

A search for high-mass resonances decaying into tau(+)tau(-) is performed using a data sample of pp collisions at root s = 7 TeV. The data were collected with the CMS detector at the LHC and correspond to an integrated luminosity of 4.9 fb(-1). The number of observed events is in agreement with the standard model prediction. An upper limit on the product of the resonance cross section and branching fraction into tau-lepton pairs is calculated as a function of the resonance mass. Using the sequential standard model resonance Z'(SSM) and the superstring-inspired E-6 model with resonance Z'(psi) as benchmarks, resonances with standard model couplings with masses below 1.4 and 1.1 TeV, respectively, are excluded at 95% confidence level. Glucocorticoids Modulate the mTOR Pathway in the Hippocampus: Differential Effects Depending on Stress History

Glucocorticoid (GC) hormones, released by the adrenals in response to stress, are key regulators of neuronal plasticity. In the brain, the hippocampus is a major target of GC, with abundant expression of the GC receptor. GC differentially affect the hippocampal transcriptome and consequently neuronal plasticity in a subregion-specific manner, with consequences for hippocampal information flow and memory formation. Here, we show that GC directly affect the mammalian target of rapamycin (mTOR) signaling pathway, which plays a central role in translational control and has long-lasting effects on the plasticity of specific brain circuits. We demonstrate that regulators of the mTOR pathway, DNA damage-induced transcript (DDIT) 4 and FK506-binding protein 51 are transcriptionally up-regulated by an acute GC challenge in the dentate gyrus (DG) subregion of the rat hippocampus, most likely via a GC-responseelement-drivenmechanism. Furthermore, two other mTOR pathway members, them TOR regulator DDIT4-like and the mTOR target DDIT3, are down-regulated by GC in the rat DG. Interestingly, the GC responsiveness of DDIT4 and DDIT3 was lost in animals with a recent history of chronic stress. Basal hippocampal mTOR protein levels were higher in animals exposed to chronic stress than in controls. Moreover, an acute GC challenge significantly reduced mTOR protein levels in the hippocampus of animals with a chronic stress history but not in unstressed controls. Based on these findings, we propose that direct regulation of the mTOR pathway by GC represents an important mechanism regulating neuronal plasticity in the rat DG, which changes after exposure to chronic stress. Combined Search for the Standard Model Higgs Boson Decaying to a b(b)over-bar Pair Using the Full CDF Data Set

We combine the results of searches for the standard model (SM) Higgs boson based on the full CDF Run II data set obtained from root s = 1.96 TeV p (p) over bar collisions at the Fermilab Tevatron corresponding to an integrated luminosity of 9.45 fb(-1). The searches are conducted for Higgs bosons that are produced in association with a W or Z boson, have masses in the range 90-150 GeV/c(2), and decay into b (b) over bar pairs. An excess of data is present that is inconsistent with the background prediction at the level of 2.5 standard deviations (the most significant local excess is 2.7 standard deviations).
