W-boson polarization measurement in the t(t)over-bar dilepton channel using the CDF II detector

We present a measurement of the W-boson polarization in top-quark decays in t (t) over bar events with decays to dilepton final states using data corresponding to 5.1 fb(-1) of integrated luminosity in p (p) over bar collisions collected by the CDF II detector at the Tevatron. Assuming a top-quark mass of 172.5 GeV/c(2), a simultaneous measurement of the fractions of longitudinal (f(0)) and right-handed (f(+)) W-bosons yields the results f(0) = 0.70(-0.17)(+0.18)(stat) +/- 0.06(syst) and f(+) = -0.09 +/- 0.09(stat) +/- 0:03(syst). Combining this measurement with our previous measurement based on single-lepton final states, we obtain f(0) = 0.84 +/- 0.09(stat) +/- 0.05(syst) and f(+) = -0.16 +/- 0.05(stat) +/- 0.04(syst). Search for the standard model Higgs boson produced in association with a top-quark pair in pp collisions at the LHC

A search for the standard model Higgs boson produced in association with a top-quark pair is presented using data samples corresponding to an integrated luminosity of 5.0 fb(-1) (5.1 fb-1) collected in pp collisions at the center-of-mass energy of 7 TeV (8 TeV). Events are considered where the top-quark pair decays to either one lepton+jets (t (t) over bar -> l nu q (q) over bar 'b (b) over bar) or dileptons (t (t) over bar -> l(+)nu l-nu b (b) over bar), being an electron or a muon. The search is optimized for the decay mode H -> b (b) over bar. The largest background to the t (t) over barH signal is top-quark pair production with additional jets. Artificial neural networks are used to discriminate between signal and background events. Combining the results from the 7 TeV and 8 TeV samples, the observed (expected) limit on the cross section for Higgs boson production in association with top-quark pairs for a Higgs boson mass of 125 GeV is 5.8 (5.2) times the standard model expectation.

Increased Tc22 and Treg/CD8 Ratio Contribute to Aggressive Growth of Transplant Associated Squamous Cell Carcinoma

Immune suppressed organ transplant recipients suffer increased morbidity and mortality from primary cutaneous SCC. We studied tumor microenvironment in transplant-associated SCC (TSCC), immune-competent SCC and normal skin by IHC, IF and RT-PCR on surgical discard. We determined T cell polarization in TSCC and SCC by intracellular cytokine staining of T cell crawl outs from human skin explants. We studied the effects of IL-22, an inducer of keratinocyte proliferation, on SCC proliferation in vitro. SCC and TSCC are both associated with significantly higher numbers of CD3(+) and CD8(+) T cells compared to normal skin. TSCC showed a higher proportion of Foxp3(+) T regs to CD8(+) T cells compared to SCC and a lower percentage of IFN-gamma producing CD4(+) T cells. TSCC, however, had a higher percentage of IL-22 producing CD8(+) T cells compared to SCC. TSCC showed more diffuse Ki67 and IL-22 receptor (IL-22R) expression by IHC. Participation in research involving novel sampling and study designs to identify acute HIV-1 infection among minority men who have sex with men

HIV-1 infection disproportionally affects African-American and Latino men who have sex with men (MSM). Their inclusion in biomedical and behavioral research is critical to understanding and addressing HIV vulnerability. Using focus groups, we sought to understand the perceptions related to participating in biomedical research of acute/recent HIV-1 infection (AHI) using complex sampling and data collection methods to reach this hidden group at highest risk of acquiring and transmitting HIV. Given the potential impact of AHI on HIV transmission in MSM, it is important to understand this intersection for HIV prevention, care, and treatment purposes. Chromatin remodeling and alternative splicing: Pre- and post-transcriptional regulation of the Arabidopsis circadian clock

Circadian clocks are endogenous mechanisms that translate environmental cues into temporal information to generate the 24-h rhythms in metabolism and physiology. The circadian function relies on the precise regulation of rhythmic gene expression at the core of the oscillator, which temporally modulates the genome transcriptional activity in virtually all multicellular organisms examined to date. Emerging evidence in plants suggests a highly sophisticated interplay between the circadian patterns of gene expression and the rhythmic changes in chromatin remodeling and histone modifications. Alternative precursor messenger RNA (pre-mRNA) splicing has also been recently defined as a fundamental pillar within the circadian system, providing the required plasticity and specificity for fine-tuning the circadian clock. Identification of mRNAs bound and regulated by human LIN28 proteins and molecular requirements for RNA recognition

Human LIN28A and LIN28B are RNA-binding proteins (RBPs) conserved in animals with important roles during development and stem cell reprogramming. We used Photoactivatable-Ribonucleoside-Enhanced Crosslinking and Immunoprecipitation (PAR-CLIP) in HEK293 cells and identified a largely overlapping set of similar to 3000 mRNAs at similar to 9500 sites located in the 3' UTR and CDS. In vitro and in vivo, LIN28 preferentially bound single-stranded RNA containing a uridine-rich element and one or more flanking guanosines and appeared to be able to disrupt base-pairing to access these elements when embedded in predicted secondary structure. In HEK293 cells, LIN28 protein binding mildly stabilized target mRNAs and increased protein abundance. The top targets were its own mRNAs and those of other RBPs and cell cycle regulators. Alteration of LIN28 protein levels also negatively regulated the abundance of some but not all let-7 miRNA family members, indicating sequence-specific bind!

Random convergence of olfactory inputs in the Drosophila mushroom body

The mushroom body in the fruitfly Drosophila melanogaster is an associative brain centre that translates odour representations into learned behavioural responses(1). Kenyon cells, the intrinsic neurons of the mushroom body, integrate input from olfactory glomeruli to encode odours as sparse distributed patterns of neural activity(2,3). We have developed anatomic tracing techniques to identify the glomerular origin of the inputs that converge onto 200 individual Kenyon cells. Here we show that each Kenyon cell integrates input from a different and apparently random combination of glomeruli. The glomerular inputs to individual Kenyon cells show no discernible organization with respect to their odour tuning, anatomic features or developmental origins. Moreover, different classes of Kenyon cells do not seem to preferentially integrate inputs from specific combinations of glomeruli. This organization of glomerular connections to the mushroom body could allow the fly to context!

Male circumcision decreases penile sensitivity as measured in a large cohort

Objectives To test the hypothesis that sensitivity of the foreskin is a substantial part of male penile sensitivity. To determine the effects of male circumcision on penile sensitivity in a large sample. Subjects and Methods The study aimed at a sample size of approximate to 1000 men. Given the intimate nature of the questions and the intended large sample size, the authors decided to create an online survey. Respondents were recruited by means of leaflets and advertising. Results The analysis sample consisted of 1059 uncircumcised and 310 circumcised men. For the glans penis, circumcised men reported decreased sexual pleasure and lower orgasm intensity. They also stated more effort was required to achieve orgasm, and a higher percentage of them experienced unusual sensations (burning, prickling, itching, or tingling and numbness of the glans penis). For the penile shaft a higher percentage of circumcised men described discomfort and pain, numbness and unusual sensations. In comparison to men circumcised before puberty, men circumcised during adolescence or later indicated less sexual pleasure at the glans penis, and a higher percentage of them reported discomfort or pain and unusual sensations at the penile shaft. Conclusions This study confirms the importance of the foreskin for penile sensitivity, overall sexual satisfaction, and penile functioning. Furthermore, this study shows that a higher percentage of circumcised men experience discomfort or pain and unusual sensations as compared with the uncircumcised population. Before circumcision without medical indication, adult men, and parents considering circumcision of their sons, should be informed of the importance of the foreskin in male sexuality.

Resilience and vulnerability: a neurobiological perspective.

The brain is constantly adapting to a changing environment. It detects environmental stimuli, integrates that information with internal states, and engages appropriate behavioral and physiological responses. This process of stability through change is termed "allostasis", and serves as a mechanism by which an organism can adapt to a changing environment to function optimally, and ultimately ensure survival. The ability to adapt to stressors in the environment by "bending" but not "breaking" can be considered as "resilience". Individuals that are more able to withstand such challenges to their stability, and bounce back after, can be considered more resilient than those that do not. This review will explore what resilience means in a neurobiological context, the role of stress and allostasis, and focuses on the role of neurotrophins, particularly BDNF, in mediating adaptive plasticity.

A structural role for the PHP domain in E. coli DNA polymerase III

Background: In addition to the core catalytic machinery, bacterial replicative DNA polymerases contain a Polymerase and Histidinol Phosphatase (PHP) domain whose function is not entirely understood. The PHP domains of some bacterial replicases are active metal-dependent nucleases that may play a role in proofreading. In E. coli DNA polymerase III, however, the PHP domain has lost several metal-coordinating residues and is likely to be catalytically inactive. Results: Genomic searches show that the loss of metal-coordinating residues in polymerase PHP domains is likely to have coevolved with the presence of a separate proofreading exonuclease that works with the polymerase. Although the E. coli Pol III PHP domain has lost metal-coordinating residues, the structure of the domain has been conserved to a remarkable degree when compared to that of metal-binding PHP domains. This is demonstrated by our ability to restore metal binding with only three point mutations, as confirmed by the metal-bound crystal structure of this mutant determined at 2.9 angstrom resolution. We also show that Pol III, a large multi-domain protein, unfolds cooperatively and that mutations in the degenerate metal-binding site of the PHP domain decrease the overall stability of Pol III and reduce its activity. Conclusions: While the presence of a PHP domain in replicative bacterial polymerases is strictly conserved, its ability to coordinate metals and to perform proofreading exonuclease activity is not, suggesting additional nonenzymatic roles for the domain. Our results show that the PHP domain is a major structural element in Pol III and its integrity modulates both the stability and activity of the polymerase.
