High-Throughput and Spectroscopy Resource Center
High-Throughput Screening (HTS) is an important step in the discovery of new medicines and involves the miniaturization and automation of bioassays so that millions of compounds can be tested. The High-Throughput and Spectroscopy Resource Center (HTSRC) supports researchers in improving the efficiency of their bioassays, identifying compounds and genetic modulators of function, and in utilizing core technologies typically applied to biochemical analysis. The center has a collection of 276,560 compounds, automated liquid transfer devices, compound databases, and supports a broad diversity of assay development techniques, typically found in early drug discovery programs.
For assay technologies, the HTSRC has the capability to support cellular and biochemical assays using absorbance, fluorescent kinetics, fluorescence anisotropy,time-resolved fluorescence, time-resolved fluorescence resonance energy transfer, AlphaScreen, bioluminescence, for example luciferase and green fluorescent protein, scintillation proximity, cellular fluorescence imaging and SPE-Mass Spectrometer. Assay targets can include ion channels, receptors, enzymes, protein interactions, signaling pathways and cellular processes.
The Center also supports spectroscopic and calorimetric equipment for use in studies of the structure, function and interactions of biological and organic molecules. The following instruments and associated techniques are available: a microscale thermophoresis instrument, a circular dichroism spectrometer (CD), a surface plasmon resonance detector (SPR), an isothermal calorimeter (ITC), 2 NMRs and a high throughput Dynamic Light Scattering Instrument
On a collaborative basis, the HTSRC can support projects from academic institutions outside of Rockefeller or Weill-Cornell Medical College; however, the HTSRC does not currently support research in conjunction with for-profit entities.
